唾液酸的结构特征、修饰及其分析方法的进展

摘要


唾液酸是一类在脊椎动物、某些无脊椎动物、真菌和细菌中广泛存在的9-碳羧基化单糖。唾液酸的三种主
要结构类型包括N-乙酰神经氨酸(Neu5Ac)、N-羟乙酰神经氨酸(Neu5Gc)和酮基-脱氧壬酮糖酸(KDN),它
们中的O-乙酰化是一种常见的化学修饰。用于唾液酸分析的常规策略包括1,2-二氨基-4,5-甲基二氧苯(DMB)
衍生化结合液相色谱-荧光检测或液相色谱-质谱(LC-MS),但DMB在对某些O-乙酰化唾液酸进行色谱分辨时
表现不佳。过去几十年中,科学家们已经开发了多种方法以提高唾液酸的色谱分离效率和检测灵敏度,展望未来,
这一领域还有进一步发展的趋势和需求。
