探索脂质代谢基因表达与AML预后的关系

摘要


目的:通过生物信息学方法探讨脂质代谢在急性髓系白血病(AML)发生发展中的作用,并对AML患者建
立危险分层方法,确定预后相关基因,从而为个体化靶向治疗提供可能。方法:从UCSC数据库下载AML患者的基
因表达RNAseq和临床资料并进行筛选。从MsigDB下载获得脂质代谢相关基因(LMRGs)。使用 ConensusClusterPlus
聚类法确定新的分子亚群。基因本体功能注释(GO)、京都基因与基因组百科全书(KEGG)、基因集变异分析
(GSVA)和Metascape数据库进行功能分析。Estimate算法和 CIBERSORTx确定骨髓微环境中的免疫状态和免疫细胞
相对丰度。Lasso回归和多因素 Cox回归分析建立预后风险模型。结果:本研究基于 LMRGs表达和临床数据鉴定出两
个存活率有显著差异的亚组。较好的预后与间质评分低、免疫评分低、免疫状态相对较低有关。Metascape富集分析
显示,两个亚组之间的差异表达基因(DEG)主要集中在免疫和骨重建相关的通路上。GSVA分析表明,脂质分解
代谢和脂质羟基化可能影响免疫和骨重建。基于 LMRGs构建的预后风险模型在预测AML患者生存显示出一定的准
确性。结论:LMRGs的表达与AML患者的免疫和骨重建和有关,可用于对AML患者危险分层以及预测生存。
