经环磷酸腺苷(cAMP)通路探究淫羊藿苷 对人精子冻融损伤的影响研究
摘要
精子冻融损伤的保护作用及其分子机制。方法:采用标准快速冷冻法建立人精子冻融损伤模型。实验设置新鲜对照
组、冻融对照组及在冷冻液中添加淫羊藿苷的干预组。复苏后,应用流式细胞术检测精子活性,并采用RT-qPCR
技术检测cAMP下游SIRT1/Nrf2通路相关基因的mRNA表达。结果:与新鲜对照组(活性精子比例83.00%±5.70%)
相比,冻融对照组的活性精子比例显著下降至65.00%±4.29%(P<0.0001)。在冷冻液中添加淫羊藿苷能进一步将活
性精子比例提升至73.00%±4.81%(P<0.01 vs 冻融对照组)。机制研究发现,淫羊藿苷显著上调了cAMP下游效应分
子SIRT1、NFE2L2(Nrf2)及其下游抗氧化基因(HMOX1,GPX4,SOD2,CAT)的mRNA表达。结论:不加磷
酸二酯酶抑制剂处理使活性精子比例从对照组的83.00%±5.70%显著下降至65.00%±4.29%(P<0.0001)。100 μg/mL
及以上浓度的淫羊藿苷能显著改善冷冻氧化损伤效应,其中200 μg/mL组的保护效果最强,将活性精子比例恢复至
73.00%±4.81%(P<0.0001)。淫羊藿苷处理还显著上调SIRT1、NFE2L2、HMOX1、GPX4、SOD2及CAT的mRNA
表达(P<0.0001)。
