基于网络药理学探讨锦灯笼抗白血病的作用机制
摘要
方法:借助TCMSP、Swiss Target Prediction、Drug Bank、OMIM及Genecards等数据库,预测锦灯笼的活性成分与
白血病相关疾病靶点;获取两者交集靶点后构建蛋白质-蛋白质相互作用(PPI)网络,利用String与Cytoscape3.10
分析平台绘制“成分-疾病-靶点”PPI网络图;基于Metascape分析平台对核心靶点开展GO功能富集分析与KEGG
通路富集分析;采用Pymol及Auto Dock Tools软件进行分子对接实验,模拟活性化合物与疾病靶点的结合活性。
结果:锦灯笼活性成分共44种,对应329个疾病相关靶点;白血病疾病靶点1750个,锦灯笼与白血病的交集靶点有
41个;筛选得到潜在作用通路10条,涵盖核苷酸结合、ATP结合、人乳头瘤病毒感染等;分子对接结果显示,锦灯
笼活性化合物与核心靶点具有较强的结合亲和力。结论:锦灯笼中的木犀草素或许是其治疗白血病的关键活性成分,
这一结果体现了锦灯笼多靶点协同治疗白血病的作用特征,可为后续锦灯笼的深入研究与开发应用提供参考依据。
